Lasinteraccionesqueestablecenlossegmentostransmembranaen el heterodímerosondeterminantesparalafuncionalidaddelreceptor.Enestátesisempleamos monocapasdeLangmuircomosistemademembranamodelo,ysimulacionesdedinámicamolecular en bicapaslipídicasparaestudiarlafuncióndelambientelipídicoenlaconformaciónyorientación de lossegmentostransmembranaeintracelulardelaintegrina IIb3. De estosestudiossurgieronunaseriedeideasnovedosas,entreellas:(i)lospéptidospresentan una orientaciónverticalrespectoalplanodelasmonocapasdeLangmuir;(ii)elanálisisde polaridadsugierequeelsegmentointracelularestáencontactoconlasubfaseacuosayel segmento transmembranaquedaexpuestohaciaelaire;(iii)lapresenciadelospéptidosmodifica las propiedadesreológicasdelamembranacomolatensiónsuperficial,elasticidadlateralymódulo de compresibilidad;(iv)seobservóqueelsegmentode IIbinteraccionapreferentementecon lípidos enestadolíquidoexpandido(POPColamezclaPOPC-POPG)queunlípidoenestado líquido condensado(DPPC);(v)engeneral,elambientelipídicomodulalasinteraccionesyla orientaciónentrelossegmentostransmembranadelheterodímero. Los resultadosexperimentalesybioinformáticospresentadosaquídemuestranlaimportancia de comprenderlaspropiedadesdelentornoenelqueseencuentranlasproteínasdemembranay el impactoquelanaturalezadelambientelipídicotienesobresusconformacionesyfuncionalidad. Integrins areexamplesofintegralmembraneproteins.Theyarelargeheterodimericcellsurface receptors(-) thatplayakeyroleinformingadhesioncomplexeswiththeextracellularmatrix and areinvolvedinseveralsignaltransductionpathways.Integrin IIb3 isfoundinplateletsand is theprimarymodulatorofplateletaggregation.Initsinactivestate,thetransmembranesegments of integrin IIband 3 formaheterodimerwithaspecificrelativeorientation.Thedissociationof this heterodimer,leadingtoreceptoractivation,istriggeredbythebindingofcytoplasmicligands to theintracellularsegmentof 3. Theinteractionsestablishedbythetransmembranesegments in theheterodimerarecrucialforthefunctionalityofthereceptor.Inthisthesis,weuseLangmuir monolayersasamodelmembranesystem,alongwithmoleculardynamicssimulationsinlipid bilayers,tostudytheroleofthelipidenvironmentintheconformationandorientationofthe transmembraneandintracellularsegmentsoftheintegrin IIb3. Fromthesestudies,aseriesofnovelideasemerged,including:(i)thepeptideshaveavertical orientationwithrespecttotheplaneoftheLangmuirmonolayers;(ii)Polarityanalysissuggests the intracellularsegmentisincontactwiththeaqueoussubphaseandthetransmembranesegment is exposedtotheair;(iii)thepresenceofthepeptidesalterstherheologicalpropertiesofthe membrane,suchassurfacetension,lateralelasticity,andcompressibilitymodulus;(iv)itwas observed thatlipidssuchasPOPCandPCPGpreferentiallyinteractwiththe IIbpeptide;and (v) ingeneral,thelipidenvironmentmodulatestheinteractionsandorientationbetweenthe transmembranesegmentsoftheheterodimer.. The experimentalandbioinformaticsresultspresentedheredemonstratetheimportanceof understanding thepropertiesoftheenvironmentinwhichmembraneproteinsarefound,aswellas the impactthatthenatureofthelipidenvironmenthasontheirconformationsandfunctionality. |
